When I first managed salespeople, I was laser-focused on activity. Over time I learned that, even though an active rep will bump into deals, you can’t build a high-performing sales organization on activity alone. That takes a careful balance of both activity and effectiveness. You have to do as much work—or more—on building that effectiveness.Hire for attributes. Even though I worked in staffing, I defaulted to always hiring for experience. That’s always my clients wanted, too. I believed that by hiring for experience, I wouldn’t have to do so much managing. I believed it would make my job easier. Over time, I found that managing is easier when you hire people with the right attributes and coach, train, and develop them.You live and die by your pipeline. It’s easy to be distracted by all kinds of sales management tasks. But the real action is in the pipeline. I wish I’d have known that your future is easily predicted in the pipeline you have right now. If you want to know what the next two quarters look like, your pipeline has the answer. And there is no cramming for sales results.Serve salespeople before the organization. The organization makes incredible demands of the sales manager. Many of those demands do nothing to help the sales manager or to help his team produce better results. The more time I spent with salespeople, actively helping them with their live deals, the better the results. Even if the organization has to wait for what it needs, I learned it is better to serve the sales force first.You sell to your organization. I had know idea how much time I would need to spend selling within my own organization. Even though we all want the same things, there are all kinds of agreements that need to be negotiated. For clients. For the sales force. For the company’s benefit. Your team needs you to sell on their behalf. And, a lot of the time, you’re the only one that can make the internal sale.You build and manage the process. It was great to find someone that could sell. It was awesome to help someone grow into an effective salesperson. But the challenge is in notching the whole sales organization up. The real key to a high performing sales force is in building and managing a process that improves the performance of the 80% of the sales force that makes the top 20% possible.You have to see for yourself. If you really want to know how a salesperson performs in front of your prospective clients, you have to go see for yourself. It’s impossibly difficult to know how to help if you haven’t seen it for yourself. My ability to know how to help salespeople was improved by seeing things for myself.There is only one forecast date that matters. It doesn’t matter what the close date in your sales force automation says, if it isn’t a date your dream client has agreed to, the opportunity isn’t going to close on that date. Period.Show no mercy when cleaning the pipeline. Salespeople are happy to work on opportunities. They need opportunities to make their number. But much of what comes into the pipeline aren’t really opportunities. I wish I’d have known sooner that you have to protect the sales force from themselves when it comes to disqualifying. Show. No. Mercy.The big lever is caring enough to personally coach. I managed salespeople. But, if you want a high performing sales force, you need to care enough about the individuals on your team to coach them directly. It takes time and energy. Everyone Thinks Justin Verlander Belongs In The Hall Of Fame So Why

Even Major League Baseball’s official Twitter account called it “ANOTHER line on Justin Verlander’s Hall of Fame résumé.” But are we all being a bit too hasty?By at least one smart measure of Hall-worthiness, Verlander is not yet deserving of a berth in Cooperstown. Cited by many sabermetrically-inclined voters, Jaffe Wins Above Replacement Score, or JAWS, is the average of a player’s career wins above replacement2JAWS uses the Baseball-Reference.com version of this stat, so I will do the same throughout this article. and his WAR over his seven best seasons (“peak WAR”). To determine if a player belongs in the Hall, a player’s JAWS is compared with the average JAWS of existing Hall of Famers at his position (because it’s easier to accrue value at, say, starting pitcher than at catcher). If the player’s JAWS is higher, the system recommends a yes vote; if the Hall of Famers’ JAWS is higher, the player falls short. Although JAWS is an imperfect yardstick (for example, it doesn’t include postseason stats, nor does it account for unique accomplishments like, say, pitching three no-hitters), electing only people above its positional standards ensures that the quality of Hall of Fame players does not decrease.With a JAWS of 59.0 so far in his career, Verlander remains below the JAWS average for starting pitchers of 61.5. But he’s not alone. No active starting pitcher meets the JAWS standard (although some may do so by the time they retire). In fact, neither does any starter who has thrown a single pitch this entire decade. That suggests a problem not with Verlander or other modern pitchers, but with the standard. Simply put, it’s too high given the usage patterns of today’s starting pitchers.3For the record, this isn’t just a problem with JAWS. Perhaps the most famous old-school standard for a Hall of Fame starting pitcher, winning 300 games, may also now be obsolete; it’s been 10 years since anyone won their 300th, and people are openly wondering if it will ever happen again.As pitch counts and bullpens have become bigger parts of the game, we’ve gone from 1,034 complete games pitched in the 1978 season to 266 in 1997 to just 42 last year. From 1871 to 1953, a period that includes at least part of the careers of about two-thirds of Hall of Fame starting pitchers, starters accounted for more than 80 percent of all innings pitched every season. In 2018, that share was 60 percent. It’s simply not fair to compare starting pitchers of the past few decades — let alone of the past few years, a period when “bullpenning” has exploded in popularity — with bygone Hall of Famers who regularly exceeded 300 innings pitched in a year.What we can do instead is sketch out a new standard, based on the premise that the Hall of Fame should immortalize the greatest starting pitchers of each era, even if those eras are not directly comparable. Verlander’s numbers may not be able to hold a candle to those of, say, the deadball era, but he deserves recognition as an elite hurler in the context of our current era, the bullpen-happy 2010s. The average season from 1901 to 20044I started with 1901 because that is the year the American League was founded and “modern” baseball (defined broadly) began; I ended with 2004 because players who were active in 2005 and later are still having their Hall of Fame candidacies considered by the Baseball Writers’ Association of America. had 10 active starting pitchers who were eventually elected to Cooperstown, so we can maintain the Hall’s relative historical standards by electing the top 10 best starting pitchers in the game today. But the No. 10 pitcher on the list, Adam Wainwright, is probably not a Hall of Famer — and his 36.7 JAWS likely shouldn’t be used as the new standard. That’s because young studs like Aaron Nola (20.2 JAWS at age 26) are still racking up stats and could eventually displace the bottom several names on the list. So let’s try looking at the version of this list from, say, 2012. It would be surprising if Chris Sale (42.3 JAWS at age 30) didn’t crack it. Madison Bumgarner (33.6 JAWS at age 30) and Stephen Strasburg (30.8 JAWS at age 31) are young enough that they could get there in time as well. Félix Hernández (44.6 JAWS at age 33) and Jon Lester (40.3 JAWS at age 35) are close but may be running out of gas. In all likelihood, though, the first several names on this list are safe — which would mean that we know the JAWS threshold for cracking the top 10. Essentially, it looks like amassing around 50 JAWS for your career is enough to guarantee your place as one of the top 10 starting pitchers of the early 2010s. And, if the trend of increased bullpen usage of the past few years continues through the next decade, the standard for the early 2020s may be even lower.This revision has a couple of implications for Hall of Fame selection that voters should heed before it’s too late. First, despite falling well short of historical standards, candidates like Johan Santana (who received very little support on the one Hall of Fame ballot he appeared on, despite currently being the eighth-best starter active in 2012) deserve closer looks, as they were elite for their era. Second, yes — Justin Verlander is a no-doubt Hall of Famer. Streak continues for No 1 Bucks

Winning 20 games in a season is nothing new to Ohio State men’s basketball coach Thad Matta. Winning 20 games before picking up his first loss, however, is a different story. OSU’s 73-68 win against Illinois on Saturday advanced the Buckeyes’ record to 20-0 and earned Matta his 11th consecutive season with 20 or more wins as a college basketball head coach. Only Mark Few of Gonzaga has started his career with such a streak. “It’s amazing,” Matta said. “You always want to get to 20. I mean, that was kind of like the benchmark for college basketball.” The Buckeyes’ 20-0 start is OSU’s first since the 1961-62 season when the team started the season with a 22-0 record. A season earlier, the 1960-61 squad got off to the best start in program history with a 27-0 record. Both of those teams went on to become the national runner-up in their respective seasons. Despite the Buckeyes’ hot start, Matta said he hasn’t noticed anyone on his team pay any attention to it. “I haven’t heard one guy say one word about not being beaten,” Matta said. “The focus has, I think, been primarily on who’s the next opponent, and you know, the job that we have to do to win that basketball game.” Kansas’ loss to Texas on Saturday — which snapped the Jayhawks’ 69-game win streak at home — leaves OSU and San Diego State as the only undefeated teams in the country. With six of their next eight games coming against ranked opponents — including road matchups at Minnesota, Wisconsin and Purdue — the Buckeyes have their work cut out for them as they attempt to remain unbeaten. “The Big Ten’s a great conference, and there’s no easy games, especially on the road,” senior guard Jon Diebler said. “We’ve got some great tests coming up, and we’re definitely looking forward to those challenges.” OSU’s next test will be when it hosts Purdue at 9 p.m. Tuesday. Andrew Holness, on Friday (February 23), handed over keys to contributors who have purchased one-bedroom units at The Estuary in Friendship, St. James.The scheme was developed under a joint venture arrangement between the National Housing Trust (NHT) and West Indies Home Contractors Limited.  When completed, The Estuary will comprise 1,500 units and supporting infrastructure on 254 acres of land.  The development is being done at a cost of $7 billion, and is targeted at low income earners.In his remarks, Mr. Holness said the NHT is on track to issue 8,000 mortgages for the 2017/18 financial year.“This is the highest it has done in any decade. You can see that the Government is being very effective in using an institution like the NHT to create housing solutions for the people,” he stated.The Prime Minister commented that the NHT was designed to promote formal home ownership in Jamaica, however “the challenge has always been to find a way to ensure that every Jamaican has access to this formal process”.“I have made it a point of duty to come to all of these ceremonies, whether it is groundbreaking or handing over, to prove the point that the Government of Jamaica can use the resources at its disposal in an efficient and effective way to achieve Government policy and to deliver benefits to the people in a timely way,” he added.Meanwhile, Mr. Holness has given the commitment that the roadway leading to the community.“There are plans already in place for the roads to be repaired and this will happen in short order. Jaguar Land Rover could use ultraviolet light to kill germs in your

That’s a lot of time in which, the company says, your car could be doing more to keep you healthy. To that end, future Jaguar and Land Rover models could use special ultraviolet light — UV-C light — in order to kill germs before they get through the car’s ventilation system.The UV-C technology, which uses ultraviolet light in the 200-280-nanometer wavelengths, has been used in the medical field to sterilize surfaces and air by neutralizing bacteria and other pathogens. JLR’s idea is to integrate this into the many hidden vents and ducts inside your car’s dashboard, so the air that gets blown into the cabin has already been sterilized. Some JLR models already use air ionization tech in their climate-control systems, but the UV-C system would be more effective.JLR says the technology could prevent the spread of common colds and flus, but also says that studies show UV-C technology can cut the transmission of drug-resistant superbugs, including MRSA, VRE, C. difficile and Acinetobacter, by 30 percent. Roush Performance Roush Performance is turning its attention to Ford pickup trucks once again, announcing on Thursday that it is launching a Roush Super Duty. 2 cops held over extortion 4 rescued

Juba League man killed in Bogra

(AP Photo/Dita Alangkara)“I almost died after giving birth to my daughter, Olympia,” Williams wrote in an essay published on CNN Tuesday.Williams, who said she had a fairly easy pregnancy, had complications in labor when her daughter’s heart rate dropped during contractions, resulting in an emergency C-section.“The surgery went smoothly.  Before I knew it, Olympia was in my arms. It was the most amazing feeling I’ve ever experienced in my life,” she wrote.However, Williams’ maternal excitement quickly turned into fear.  “But what followed just 24 hours after giving birth were six days of uncertainty,” the tennis player wrote.First, because of her medical history with this issue before, Williams was able to self-diagnose a pulmonary embolism, “a condition in which one or more arteries in the lungs become blocked by a blood clot,” she explained.The pulmonary embolism ignited more health issues and measures, including a torn C-section wound, a hematoma, which is inflammation of clotted blood, in her abdomen, and then a procedure to prevent anymore blood clots.  After she was able to return home from the health scares, she was confined to bed for over a month.“I had to spend the first six weeks of motherhood in bed,” Williams wrote.With fame and fortune, Williams had access to some of the best doctors and healthcare.  “Yet I consider myself fortunate,” she wrote.“I am so grateful I had access to such an incredible medical team of doctors and nurses at a hospital with state-of-the-art equipment.  They knew exactly how to handle this complicated turn of events.  If it weren’t for their professional care, I wouldn’t be here today,” the 36-year-old wrote.Her near death experience brought attention to a sobering reality- many women, in particularly those of color, would not have been as lucky as the rich and revered tennis player.“According to the Centers for Disease Control and Prevention, [Black] women in the United States are over three times more likely to dies from pregnancy or childbirth-related causes,” Williams wrote.Beyond the realities Black mothers in America face in pregnancy and labor, Williams also considered the thousands of women in poor countries around the world.“When they have complications like mine, there are often no drugs, health facilities or doctors to save them.  If they don’t want to give birth at home, they have to travel great distances at the height of pregnancy.  Before they even bring a new life into this world, the cards are already stacked against them,” Williams wrote.In her essay, Williams pondered a world where there was sufficient access to adequate birthing attendants, clean water, and affordable and accessible healthcare.“That world is possible.  And we must dare to dream it for every Black woman, for every woman in Malawi, and for every mother out there,” she wrote.last_img read more

Nanocontainers for nanocargo Delivering genes and proteins for cellular imaging genetic medicine

first_imgIn their paper, the authors report that the T4 capsid NP gene expression and protein delivery system may be complementary to or used in conjunction with gene therapy based on RNA Cas and taran nuclease. (Cas genes code for proteins related to DNA loci containing short repetitions of base sequences known as Clustered Regularly Interspaced Short Palindromic Repeats, or CRISPRs.) “The T4 nanoparticle expression system can easily complement Cas9 and taran nuclease-based recombination by packaging the linear cas9, target-sgRNA plasmid DNA, and Cre recombinase – or even ligase, an enzyme that facilitates the joining of DNA strands – and deliver the resulting T4 nanoparticles into the recipient eukaryotic cells with high specificity employing SOC and HOC,” Liu tells Phys.org. (SOC and HOC are dispensable T4 capsid proteins.) “By displaying the targeting ligands (binding molecules) onto the surface, the T4 capsid gene expression and protein system will be able to efficiently deliver the Cas9 and sgRNA plasmids together into the desired recipient cells. Relevant enzymatically-active proteins Cas9, lambda exonuclease, lambda beta protein and others can be delivered directly at the same time from the T4 nanoparticle.” Seamless gene correction of beta-thalassemia mutations in patient-specific cells Dr. Jinny L. Liu discussed the paper that she, Prof. Lindsay W. Black and their co-authors published in Proceedings of the National Academy of Sciences USA. “Icosahedral viral nanoparticles are essentially 100 nm by 80 nm nanocontainers that allow exogenous genetic material to be packaged in vitro through nucleic acid machinery that generally only allows linear DNA/RNA to be packaged through a portal channel,” Liu tells Phys.org. “However, in vitro protein packaging is generally impossible, because for most viral nanoparticles there is no protein packaging machinery comparable to nucleic acid packaging machinery.” While protein may be chemically cross-linked to the capsid inner surface, this is expected to lead to protein denaturation and loss of enzymatic activity.That being said, nature has evolved solutions to this protein packaging conundrum. During in vivo viral capsid assembly, Liu explains, some bacterial viruses, or bacteriophages, target proteins within the procapsids before the nucleic acid is packaged so as to eject the proteins with the nucleic acid, thereby facilitating infection in conjunction with the nucleic acid. (A procapsid, or prohead, is an immature viral capsid structure formed in the early stages of self-assembly of some bacteriophages. Production and assembly of stable proheads is an essential precursor to bacteriophage genome packaging.) Only a few phages have well-characterized in vivo protein packaging systems, and phage T4 is the best characterized. “Prof. Black’s lab at UMB and my lab at NRL have demonstrated that not only can a specific foreign enzyme – cyclic recombination (Cre) recombinase – be packaged into the capsid in vivo, but also that it is active within the capsid.” This activity was demonstrated by showing the religation (the rejoining of two DNA strands or other molecules by a phosphate ester linkage) of packaged linear DNA flanked with two Cre recombination sites.The paper shows that the substantial space within a T4 nanocontainer accommodates the active Cre enzyme along with exogenous DNA. “For potential applications, T4 can package up to 50 kb exogenous linear DNA containing full-length desired genes along with recombinases, either Cre or λ-red proteins, for specific homologous recombination within the chromosome,” Liu notes. (Homologous recombination is a type of genetic recombination in which nucleotide sequences are exchanged between two similar or identical molecules of DNA.) “We expect that the cas9 enzyme could be encapsidated in a comparable way – and in fact, at least eight different proteins have been encapsidated in this manner. Through homologous recombination, our system can allow the corrected gene to replace the mutated gene in its original location within the chromosome or by precisely knocking out the overactive genes in stem cells.” Liu points out that the T4 delivery vector is safer and better controlled than other viral delivery gene therapy, such as those delivering genes using infectious animal viral vectors to randomly insert the gene within the chromosome. Liu adds that her lab has also been studying cell imaging and drug/gene delivery to eukaryotic cells using T4 tailless nanoparticles, which the researchers demonstrated can enter the eukaryotic cells without causing cell death.A specific example of potential downstream drug and gene therapeutic applications resulting from the new approach is delivery of the toxic protein and linear plasmid that produces neutralizing peptides or antibodies into targeted cancer cells displaying specific cancer markers using high affinity SOC + HOC marker binding proteins on the surface of the capsids, while another example is to use the system for HIV gene therapy. Liu adds that there are several pathways to use this system for gene therapy:Delivering T4 nanoparticles packaged with the recombinase (or ligase) and linear plasmid DNA to produce gp120 or interferon to generate or boost the immune response in patientsDelivering T4 nanoparticles packaged with recombinase (ligase) and the linear soluble CD40 expression plasmid DNA into T lymphocytes or hematopoietic cells to block the infection of HIV-1Inhibiting RNA by delivering the engineered plasmid DNA that can produce decoy RNA for binding the viral sense DNAInhibiting protein by delivering packaged anti-viral antibodies and anti-HIV antibody plasmid DNA In addition to diagnostic and cellular imaging, the T4 nanoparticle gene-protein system can deliver repaired genes to correct human genetic diseases – for example, reversing adenosine deaminase (ADA) deficiency by introducing the protein-DNA complex to express ADA in stem cells. Other broad areas of research impacted by gene therapy technologies, such as genetic defects, cancer, neurological diseases in adults, and aging itself, may also benefit from this study.Moving forward, the scientists want to develop more T4 procapsids packaging exonuclease and other recombinases along with engineered target DNA to demonstrate that the resulting T4 capsids can insert the gene into a stem cell line with a genetic deficiency. “In addition,” Liu concludes, “we’re working on adapting our system to deliver therapeutic peptides or antibodies to cells exposed to or infected by biothreat agents, such as protein toxins or viruses, efficient neutralization of toxin effects. Discovery’s free-to-air German channels DMAX and TLC will be included.7TV exists already as P7S1’s mobile offering and has registered six million downloads since 2014.The new version of the service will utilise the same app and underlying tech, but the expanded streaming service will have a wider selection of content. Discovery and P7S1 added that they would “welcome talks with other content providers to participate”.P7S1’s SevenOne Media will sell the new service’s ad inventory when it launches later this year.
